Mother Brain is the main antagonist of the Metroid franchise, having first appeared in the game of the same name. Mother Brain has made cameos and a few mentions in Mario-related media, in which she is first mentioned in the Nintendo Comics System comic "Duh Stoopid Bomb!," whereas Mother Brain's first physical appearance in a Mario-related game is WarioWare, Inc.: Mega Microgame$!.
